CD20 is a transmembrane phosphoprotein belonging to the MS4A protein family. Its expression is highly specific to B lymphocytes, appearing first at the pre-B cell stage and persisting through to mature B cells, but is eventually lost when these cells differentiate into plasma cells. Functionally, CD20 plays a critical role in regulating B cell activation, proliferation, and differentiation. It achieves this by modulating calcium ion influx across the cell membrane and participating in B-cell receptor (BCR) signaling pathways, processes that ultimately influence cell cycle progression and antibody production. These unique characteristics—restricted expression and key regulatory functions—have made CD20 one of the most successful targets for antibody-based therapies in immuno-oncology.
