CD36, also known as fatty acid translocase, is a transmembrane glycoprotein belonging to the class B scavenger receptor family. It is widely expressed in various cell types, including macrophages, adipocytes, hepatocytes, cardiomyocytes, and platelets. CD36 functions as a multifunctional receptor involved in long-chain fatty acid transport, oxidized low-density lipoprotein (oxLDL) uptake, clearance of apoptotic cells, and anti-angiogenic activity via thrombospondin-1 (TSP-1) binding. In cancer, CD36 plays a context-dependent dual role: it promotes metastasis, stemness, and therapy resistance in most solid tumors, but acts as a tumor suppressor in triple-negative breast cancer by inducing ferroptosis. CD36 is also implicated in metabolic diseases such as non-alcoholic fatty liver disease and atherosclerosis, as well as in infectious diseases including malaria. Due to its central role in lipid metabolism, immune regulation, and tumor progression, CD36 represents a promising but challenging therapeutic target.
