CD28 is a transmembrane glycoprotein expressed on the surface of T lymphocytes. It belongs to the immunoglobulin superfamily and functions as a homodimer with a molecular weight of approximately 44 kDa per subunit. CD28 is the major costimulatory molecule for T cell activation. T cells require two signals for full activation: the first signal comes from the T cell receptor (TCR) recognizing antigen, and the second signal is provided by CD28 binding to its ligands CD80 (B7-1) and CD86 (B7-2) on antigen-presenting cells. CD28 is also essential for the development of regulatory T cells (Tregs). Due to its central role in T cell immunity, CD28 has become an important target for treating autoimmune diseases, preventing transplant rejection, and enhancing cancer immunotherapy.
